A Simple Assessment Method of Red Blood Cell Fragility

Abstract
This study proposes a simple optical method for assessing red blood cell (RBC) fragility. A sodium citrate-added porcine blood was centrifuged. The RBCs contained in the upper and lower 3% of the RBC layer were collected and each sample of 1.5μL was added to saline of 0.75mL. Here we call these samples U and L groups, respectively. Each sample was added to distilled water of 0.75mL to expose the RBCs to low osmolality. The time variation in the optical power transmitted through the sample was approximated with a first order lag function and the time constant was calculated. The mean time constants of U and L groups were 20.5s and 16.3s, respectively. The difference of the time constant seems to reflect the mean fragility of RBCs contained in the samples. Furthermore, the change in the volume distribution of RBCs was also considered through the frequency analysis of the output signal.
