Selective medium for isolating Ewingella americana, the causative pathogen of bacterial brown rot on Lentinula edodes fruit bodies

Abstract

A selective medium, designated as A-D3 medium, was developed for the isolation of Ewingella americana, the causative agent of bacterial brown rot found on the of fruit bodies of Lentinula edodes. A-D3 medium contained 10 g of D(+) arabitol, 5 g of casamino acid(vitamin free), 7 g of LiCl, 5 g of NaCl, 0.3 g of MgSO_4・7H_2O, 50 mg of sodium dodecyl sulfate, 15 mL of 0.2% bromothymol blue solution and 15 g of agar in 1,000 mL of distilled water and HCL was used to adjust the pH to 7.2. All tested isolates of Ew. americana grew well and developed characteristic colonies on the A-D3 medium, while the growth of another plant pathogenic Erwinia spp. was inhibited. The plating efficiency of Ew. americana on the A-D3 medium was 60-76% of nutrient agar medium. Using the A-D3 medium, Ew. americana was selectively isolated from the fruit bodies of L. edodes with naturally occurring brown rot symptoms.
