Flagellar Helical Parameters of Salmonella Typhimurium Observed by Laser Dark-Field Microscopy

Bacteria swim by rotating their helical flagellar filaments, which act as screw propellers. The exact shape of the filament, especially during its rotation, has been difficult to be observed because of its small size and fast rotation. In this paper, we describe a new application of laser dark-field microscopy to obtain the helical parameters of the flagellar filament of Salmonella enterica serovar Typhimurium. In one-sided laser illumination at an angle of 45° or 90°, the image of a helical flagellar filament appears as a series of spots with one or two spots per pitch of the helix, respectively. The bright spots were recorded with a high-sensitivity CCD camera equipped with an electrical shutter. The exact values of helical pitch and radius of the detached (non-rotating) flagellar filament were determined by measuring the intervals of the spots. The pitch of rotating filament on a cell was also measured. Small but significant decrease in pitch during rotation was observed.<br>
