Alteration of the activity of the β-adrenergic receptor pathway was investigated in rats with chronic myocardial infarction. Cardiac β-adrenergic receptor density (Bmax) and β-adrenoceptoradenylate cyclase activity (cAMP production in the cardiac membrane with isoproterenol-stimulation) were measured. The ratio of left ventricular weight to body weight was increased one week after myocardial infarction. Left ventricular end-diastolic pressure and heart rate increased from the 1st week and had further increased by the 10th week. The density ofβ-adrenergic receptors increased from the 1st week, but had diminished by 10th week. Both basal and isoproterenol-stimulated cAMP production in the cardiac membrane decreased from the 1st week. These results indicate that heart failure is compensated for from the 1st to the6th week of chronic myocardial infarction, but not at the 10th week. Elevation of theβ-adrenergic receptor density seems to be one of the most important compensatory mechanisms in the chronic heart failure.
