Some staining methods were introduced for basophil leucocytes. With the benzidine peroxidase reaction following the application of diluted solution of hydrochloric acid, the basophil granules of newts were stained clearly blue, although the reaction of neutrophil leucocytes contrariwise changed into negative. This finding of basophil granules was peculiar to the blood of the newt and could not be found in any other animals.<BR>Victoria blue 4R was applicable to staining of basophil granules according to the following methods: In supravital staining with its 0.01 % alcoholic solution, the newt's basophil granules were stained bluish purple, whereas they were not with Janus green B. The basophil granules of man and all the animals examined on blood smears, were stained very distinctly with aqueous solution of Victoria blue 4R in which case oxalic acid and tartaric acid were found to be an excellent mordant for it.<BR>It is worthy of note that Victoria blue 4R solution possesses an intense destructive power against blood cells of smear preparation, and for this reason these blood cells should always be fixed with formaldehyde gas sufficiently.<BR>The author wishes to express sincere thanks to Professor Mitsui at our University and to Professor Seki at Okayama University for their kind help and advice.
