- ゼブラフィッシュを用いた生殖発生毒性評価試験
Description
生殖発生毒性試験は,化合物の催奇形性を評価する試験の一つである.化合物の催奇形性の判断には,ガイドライン(ICH-S5)上では,化合物の投与期間を着床から器官形成終了時期までの間として実施する「胚・胎児発生に関する試験」が規定されている.一方,本試験は,化合物が大量に必要であること,費用が高額であること,試験が長期間になることから,創薬初期からスクリーニング的に実施するには困難な試験系である.また,近年動物削減の動きが活発になっており,生殖発生毒性試験の代替法の構築が活発に行われている.代替法として,マウスEST試験,全胚培養系などと共に,ゼブラフィッシュを用いた生殖発生毒性試験の可能性が近年注目されている.そこで,本研究では,ゼブラフィッシュを用いた生殖発生毒性評価試験を実施し,本試験系の予測性を判断した.その結果,その正答率は69%であること,本結果に他の予測試験結果を併用することで更に正答率が上昇することが明らかになった.以上から,本系の有用性が示され,創薬初期からスクリーニング的に実施することで,化合物の催奇形性リスクを早期に検出し,リスクの低い化合物を創製できる可能性があると考えられた.
