Functional Analysis of <i>Arabidopsis</i> POT1 Protein with Novel Repeats

Abstract

Postembryonic development of plant is mainly depend on the activity of meristems located at both root and shoot apex. We have identified Arabidopsis tonsoku mutants which showed the short roots and fasciated stems, suggesting the defects in meristem structure and function. Since TONSOKU protein contained the motifs used for protein-protein interaction, it seemed to be important to identify the partner of TONSOKU. POT1 was screened by yeast two hybrid, and was reveled to consist of novel repeats containing EFE core. The cellular localization of POT1 was analyzed in tobacco BY-2 cells and the protein localized at endomembrane system especially at nuclear envelope in interphase. Whereas, POT1 seemed to be localized to spindle or spindle pole body in cell division phase. These result suggest that POT1 functions in nuclear envelope and/or spindle pole body.
