Abstract

For fundamental and applied studies of tomato, sequence information such as for genomic DNA, full-length cDNA and Expressed Sequence Tags (EST), DNA markers, mutant population lines and a large collection of wild tomato species become more widely available recently. With a wealth of such information and resources, we are working on tomato breeding to obtain cultivars which are rich in functional ingredients, especially naringenin chalcone, a precursor of anthocyanins. As the first step, we made F 2 population of a Micro-Tom x LA1996, which has a locus for high anthocyanin accumulation, the Anthoyanin fruit (Aft), and scored them for anthocyanin accumulation. To genotype the F2 plants with DNA markers, we searched polymorphisms between the parental lines using DNA fragments which were PCR-amplified based on the sequences of Conserved Ortholog Set II DNA markers and anthocyanin synthesis genes, and found a DNA marker which was tightly linked to the Aft locus.
