Unraveling transcriptional regulatory network in Arabidopsis by improved yeast one-hybrid screening

Abstract

To identify transcription factors (TFs) that interact with upstream region of a gene, yeast one-hybrid screening (Y1H) is often employed. We constructed artificial semi-uniform cDNA library that only contains TF genes of Arabidopsis and showed that the library enabled to identify TF that binds to specific promoter in highly efficient manner similar as shown in nematoda. We are now trying to reveal transcriptional regulatory network of secondary wall formation that consists of ca. 100 genes using this library. Firstly, we employed 32 putative TFs involving in the secondary wall formation to examine their binding to the possible target genes one by one, and identified many binding events from more than 2,000 tests. Most of them were the regulations by the master regulators, namely, VND7 and NST1. These results suggest that these master regulators, as suggested by other studies, directly regulate various genes such as one for enzyme which located in the bottom of transcriptional regulatory cascade. We are now attempting to comprehensive analysis using all available TFs as prey and employing other region as bait to find the regulatory relations.
