2-Ethynylanilines are one of the most useful building-blocks for preparation of nitrogen-containing heterocycles, such as indoles and quinolines. We have already reported that a sort of terminal substituent on the triple bond on a 2-ethynylaniline selectively produces indoles and quinolines.<BR> Ongoing our research on the line, we have found that a catalytic amount (10 mol%) of CuBr2 promoted intermolecular cyclization of 2-ethynylanilines with N,O-acetals to produce a variety of quinoline derivatives in practical yields. These results showed a methine carbon on the N,O-acetal was embedded as a new carbon moiety in the quinoline skeleton. We have also developed a single-step synthesis of the quinoline derivative by the reaction of the 2-ethynylaniline with ethyl glyoxylate in the presence of piperidine and CuBr2.
