Expression of p53 and 53BP1 in bystander cells in the irradiation with X-ray microbeam

Description
Expression of p53 and 53PB1 was examined in the neighboring cells (bystander cells) around irradiated target cells with X-ray microbeam, which was prepared from micro-capillary equipped in the apparatus for X-ray fluorescence analysis operated at 30keV (XGT-2700 from HORIBA, Ltd.) and the microbeam irradiation system of aluminum K-shell X rays developed by Central Research Institute of Electric Power Industry (CRIEPI). The beam diameters available in each system are 100 micron and about 2 micron, respectively. The intracellular distributions of p53 and 53BP1 were detected in human lung cancer A549 cells by the immunostaining and observed under fluorescence microscope. Using XGT-2700 the defined area composed of several tens of cells was irradiated and the expression of p53 was observed outside the area after irradiation, indicating the existence of gap junction independent pathway of bystander signals. On the other hand the extended expression of 53BP1 over the target area was very small, suggesting the low incidence of DNA double strand breaks in the bystander cells. The irradiation restricted to nuclei by the CRIEPI system also confirmed the extension of p53 expression area over irradiated cells.
