Adult male rats (175g : mean body wt of 13 heads) were divided into 4 groups. Group 1 was fed basal diet devoid of vitamin B_<12> and folic acid. Groups 2 and 3 were maintained on this diet for a period of 3 weeks and then received, in treatment, methyl-B_<12> (group 2 ; intramusclar injection of 12O0μg/head/11 days) and DBCC (group 3 ; 1200μg/head/11 days). Group 4 was maintained on normal diet. All groups were sacrified and separated spinal cord of L-2,3 level and liver. After fixation, the slice in 5μ thickness was utilized for the determination of RNA and DNA contents on microspectrophotometry. Other portion of spinal cord and liver was used for the incorporation of ^3H-uridine on microradioautograph and liquid scintillation counter. RNA of motor neuron on deficient rat was less contained in spectrum than the RNA in group 4 on normal diet. Incorporation of ^3H-uridine in neurons on deficient rat was also lowered. RNA contents of motor neuron on group 2 and 3 were elevated near to the value of normal diet, and also incorporation of ^3H-uridine on both were increased, but no significant difference between group 2 and group 3. The differences of DNA in motor neurons have not been found remarkably on each group. The incorporation of ^3H-uridine in liver was elevated in groups 2 and 3 than that in group 1. It is assumed from above results that vitamin B_<12> are likely to be perfoming a significant role in maintenance and biosynthesis of RNA in neuron.
