Description
Regenerative medicine attracts attention as cutting-edge medical technology. One of the problems is how to separate the cultured cells from the culture scaffold without damage. The purpose of this study is to produce an easily dislodged cultured cell applying microstructure on the scaffold. A die was fabricated by etching Si substrate through a mask of self-assembled silica particles of which diameter were 1 μm or 5 μm. To make the environment similar to in vivo considering the oxygen permeability, the structure was copied to polydimethylsiloxane scaffold The adhesion force of cells after cultivation was measured directly peering off the cells with an AFM cantilever set on a micromanipulator. The deflection of the cantilever when the cell peered off from the scaffold was measured from the captured image and peering force was calculated. It was found that adhesion force can be decreased to 164 μN while that on flat media was 306 μN.
