The partitioning of 2-(p-toluidinyl) naphthalene-6-sulfonate (TNS) between bovine red blood cell membrane (ghost) and the bulk aqueous phase was measured by means of fluorescence spectroscopy. The negatively charged hydrophobic probe, TNS, partitions the lipid surface of red blood cell membrane. The addition of simple salt to the cell suspension resulted in an enhancement of the TNS binding. The enzymic cleavage of sialic acid residues from the glycoprotein of cell membrane also gave rise to a considerable effect on the TNS binding. The partition equilibrium of simple salt between the cell membrane and the bulk solution was examined by the use of Donnan equilibrium theory. The sequence of cation partitioning to the cell membrane was estimated to be as Ca2+>Mg2+>Li+>Na+≒K+>TMA+ (tetramethylammonium). The sequence was in agreement with the stabilizing efficiencies of cation against the glycerol hemolysis.
