The antitussive activity and site of action of 2-(2-diethylaminoethoxy) ethyl α, α-diethylphenylacetate (Oxeladin) were investigated. The 50% antitussive doses (AtD50) of Oxeladin, administered intravenously, evaluated by the "coughing dog and cat" methods, were 19.4 (16.0∼23.5) mg./kg. in unanesthetized dogs and 15.8 (13.0∼19.3) mg./kg. in pentobarbitalized cats. Compared with the AtD50 of codeine, the antitussive activity of Oxeladin is 22% and 18% that of codeine in dogs and cats, respectively, The site of action of the drug seemed to be at the cough center. This conclusion was derived from the analyses of actions as follows : (1) Comparison of equi-active doses in four different routes of administration. (2) Effect on sensory receptor in the tracheal mucosa. (3) Effect on the pulmonary stretch receptor impulses. (4)The influence of decerebration by mid-collicular transection. (5) Effects on the centrally-induced cough response and on the centrally-induced sustained inspiratory response. (6) Effect on the efferent respiratory pathways. In addition to the above results, the drug showed a weak papaverine-like action on the tracheal muscles.
