Description
The combined action of ampicillin (ABPC) and dibekacin (DKB) against Escherichia coli was bacteriologically evaluated. The results are as follows.<BR>The synergic action of ABPC and DKB was observed remarkably on Escherichia coli No.29 by chequer board titration method.<BR>The simultaneous administration of ABPC and DKB showed the highest bactericidal action, followed by the case of DKB after adding ABPC, and the reverse case showed the similar to that of each drug alone.<BR>The scanning electron microscopic observation revealed that the cell exposed to ABPC alone is extremely prolonged in a filamentous form; the cell exposed to DKB alone has the damage at the surface layer; the cell exposed to ABPC and DKB simultaneously is not prolonged in form, but has the damage at the surface layer, and the degree of surface damage is much severer than that by each drug alone.<BR>It was observed that the simultaneous administration of ABPC and DKB, in the therapeutic experiment of infection in mice, than the case of administered of DKB after administered ABPC and the reverse case.<BR>The peak concentration of ABPC and DKB in serum of mice administered subcutaneously 1mg/ mouse of ABPC and 0.8mg/ mouse of DKB attained 15 minutes after administration. The serum level of DKB maintained two times higher than that of ABPC during the experimental period.
