Abstract

6059-S, a newly developed injectable antibiotic, was administered to healthy adult volunteers to evahrate its clinical pharmacology.<BR>6059-S, when administered by intramuscular and intravenous injection or intravenous drip infusion, showed good dose-response relationship in peak blood levels respectively, and then gradually decreased with the half-life of about 2 hours.<BR>Almost all of 6059-S administered was excreted to urine in 24 hours. The renal clearance of 6059-S was about 70ml/min./1.48m2.<BR>When 6059-S was administered intravenously at the dosage of 1g, every 12 hours for 9 times, there observed no tendency of accumulation of the drug in blood or urine.<BR>Thin-layer chromatographic bioautography was conducted with the plasma and urine samples obtained from subjects administered 6059-S by intravenous injection, and no metabolite of the drug was detected from the plasma and the urine.
