The mutagenicity of AM-715, a new antibacterial drug, was tested in vitro in various short-term assays, i. e., rec-Assay, AMES test and chromosomal aberration test of cultured mammalian cells.<BR>1. rec-Assay: The ability of AM-715 preferentially to inhibit the growth of DNA-repair-deficient bacteria was weaker than that of nalidixic acid.<BR>2. AMES test: AM-715 exhibited no demonstrable mutagenicity for S. typhimurium and E. coil in the absence or presence of the microsomal preparation (S-9) derived from rat livers.<BR>3. Cytotoxicity against Chinese hamster cells (CHL cells): The growth inhibition of the cells was not observed in treatment at 100μg/ml of AM-715 for 24 hours, but was observed at 300μg/ml. Whereas, no morphological change was observed on the cells of which the growth was inhibited by treatment at 300μg/ml for 48 hours.<BR>4. Chromosomal aberration test on CHL cells: The chromosomal aberrations of the cells treated at 200μg/ml of AM-715 with or without S-9 for 48 hours were not significantly different from those of untreated cells.<BR>5. Sister chromatid exchange (SCE) in human blood cells and Chinese hamster cells (Don cells): The signiificant increase in SCE was not observed on both cells treated with AM-715.<BR>Therefore it was suggested that AM-715 had no mutagenicity in mammalian cells.
