Using β-galactosidase as label, a microtitre plate enzyme immunoassay using a double antibody method was studied for measurement of progesterone in skim milk of cows. Since standard curves were changed by presence of skim milk in the assay system, process standard curves were made by using skim milk of estrous cows containing low levels of progesterone. The sensitivity of the assay was 50 pg per well, and mean inter-and intra-assay coefficients of variation were 19.6 and 19.2%, respectively. The values measured by this assay were highly correlated (γ=0.94, n=11) with the values by radioimmunoassay for the same samples. The accuracy in assessment of pregnancy status by measurement of progesterone in skim milk of 80 cows collected at 21 to 24 days after artificial insemination, which was later confirmed by rectal palpation, was 84.2% for pregnancy and 100% for nonpregnancy.
