The formation of the zona pellucida was studied histologically in relation to the oocyte growth in the mouse ovaries from at birth till 6 days after birth. No zona pellucida was found around the oocytes at birth. Patches of the zona pellucida were initially formed between the growing oocyte and granulosa cells in the ovaries of 2-day-old mice. At 4-day, some growing oocytes became surrounded by continuous zona pellucida. The number of oocytes with such continuous zona pellucida increased until 6-day.<BR>The formation of the zona pellucida was studied in more detail using the image analyzer in relation to the diameter of oocyte, and the number and shape of the granulosa cells around the oocyte. Patches of the zona pellucida appeared around the oocyte with 33μm in diameter, and a continuous layer of zona pellucida was formed around the oocyte over 39 μm in diameter. The granulosa cells around the oocyte with discontinuous zona pellucida assumed a flat or cuboidal shape, whereas almost all granulosa cells associated with continuous zona pellucida (diameter ?? 39 μm) attained a cuboidal shape. The number of granulosa cells, thereafter, increased in proportion to the increase in oocyte diameter.
