The distribution and fluctuation of the activities of 3β-and 17β-hydroxy-steroid dehydrogenases (HSD) were studied in the ovary of IVCS mice with 4-day cycles. 1) 3β-HSD. The 3β-HSD reactions with pregnenolone or DHA as the substrate were equal. A weak to moderate 3β-HSD activity was present in the theca interna of both preantral follicles with four or more granulosa layers and of small antral follicles but none in the granulosa cells. The 3β-HSD activity appeared in the granulosa cells of large antral follicles and increased to moderate value at proestrus and estrus. The theca interna of large antral follicles developed the activity, attaining a strong activity at diestrus, proestrus and estrus. The 3β-HSD activity in the newly formed corpus luteum was greater at metestrus-2 than that at the other stages, indicating an increased function of the corpus luteum. The interstitial tissue contained a very strong activity throughout the cycle. These findings suggest histochemically that the interstitial tissue, theca interna of antral follicles and corpus luteum are likely to contribute to progesterone synthesis in the mouse ovary. 2) 17β-HSD. The enzyme activity was absent, except that a weak activity was observed in the corpus luteum and interstitial tissue in some cases.
