Genetic analysis of DNA integrated into a male founder transgenic mouse, λE1A/1, showed that about a half (44/91) of the progeny in the first generation (G1) were transgenic, and that one-fourth (12/44) of the transgenic mice had copy numbers higher than that of the founder and the rest (32/44) did lower. The ratio of the high copy numbers, the low copy numbers and the negatives was approximately 1:3:4. When G1 transgenic mice were mated with normal mice, both the high and the low copy number mice transmitted their incorporated DNA to about half of the next gen-eration without significant changes of the copy numbers. These results suggested that the founder mouse, λE1A/1, was a mosaic containing two types of transgenic cells which were produced by events at two different cleavage stages after the pronuclear injection.
