A Study on Applicability of Six Organic Solvents for Subject Chemicals to in vitro Cytotoxicity Assays

Cytotoxicity of six organic solvents, EtOH, MeOH, DMSO, FA, DMFA and EA, was examined in order to clarify their applicability to cytotoxicity assays and their allowable concentration in culture medium with use of HeLa cell culture system. The cell suspension was mixed with a sample solution, and the cells were exposed to the organic solvent in the concentration from 1.0μl/ml to 20.0μl/ml during 24 hour cultivation. After observing cells through a phase contrast microscope and counting viable cells, percentage inhibition was calculated on the basis of the ratio of normal nuclei in sample to those in control. Fifty percent inhibitive concentration (IC_<50>) was below 1.0μl/ml for EA, 4.0μl/ml for DMFA, 8.0μl/ml for EtOH, 10.0μl/ml for FA, 13.0μl/ml for MeOH and 14.0μl/ml for DMSO. The cytotoxicity potency ratio derived was DMFA : EtOH : FA : MeOH=10 : 5 : 4 : 3. EA and DMFA cannot be adequate solvents for in vitro cytotoxicity assays, by reasons of their serious cytotoxicity and their chemical properties to corrode plastics. Although DMSO showed the least cytotoxicity of the solvents tested, it was toxic enough to give considerable inhibition in the concentration above 10.0μl/ml. The foregoing results indicate that both MeOH and FA can be toxic as or less toxic than DMSO, as far as they are applied in such a range of concentration as allowable for DMSO, and that EtOH also would be applicable in a little lower concentration than that for DMSO.
