Abstract

A phylogenetic study of cultivars of Prunus mume SIEB. et ZUCC. based on isozyme variation was made. This included the detectability of isozymes, seasonal changes in their activities, and the isozyme variations in 9 cultivars. The 12 enzymes : shikimate dehydro-genase (ShDH), glycerate dehydrogenase (G 2 D), malate dehydrogenase (MDH), 6-phosphogluconate dehydrogenase (6 PG), glucose-6-phosphate dehydrogenase (G 6 P), glutamate dehydrogenase (GDH), peroxidase (POD), tetrazolium oxidase (TZO), glutamate oxaloacetate transaminase (GOT), esterase (Est), acid phos-phatase (Acp), leucine aminopeptidase (Lap) were detected with 3 types of seasonal changes in their activities. POD activity steadily increased from spring to fall, MDH, TZO, and Acp had constant activity, and the remaining 8 enzymes had two peaks of the activity, in spring and in fall. All enzymes, except MDH and Lap, were polymorphic. The 6 enzymes, ShDH, 6 PG, G6P, POD, TZO, and Est, were detected with enough repeatability, and their data were used for quantas 3 analysis. We found a good number of cultivars with double flowers. We think that is effective to collect leaves in spring and in fall for the isozyme analysis of this species.
