Effects of plant hormones and silverion treatments on <i>in vitro</i> multiple plantlet regeneration from juvenile kunugi (<i>Quercus acutissima</i>) seedlings

Abstract

The effects of the concentration of BAP (6-benzylaminopurine) and silver-ion treat-ments on the sprouting of axillary buds and the elongation of shoots were studied. The most satisfactory result in hastening the sprouting of axillary buds and elonga-tion of shoots was obtained by adding 0.8mg/l of BAP to IS (IDE and SAITO) medium. It was recognized that leaves developed swiftly after soaking the ends of the explants in 1, 000 ppm of liquid silver nitrate (AgNO3) solution for 3 seconds. The effects of supplements of IBA (3-indolebutylic acid) and NAA (α-naphtylacetic acid) and the period of STS (silver thiosulphate) treatment on root-ing were studied. Rooting of 82% was achieved by the supplement of IBA at 0.5 mg/l and NAA at 0.0002mg/l to the rooting medium. Rooting was improved by soaking the ends of the shoots for 5 seconds in STS solution before transplant-ing them to the rooting medium. Thus it was proved that silver-ion treatments are effective for in vitro multiple plantlet regeneration from juvenile kunugi (Quercus acutissima CARR.) seedlings.
