Abstract

In vitro plantlet regeneration of konara(Quercus serrata THUNB.) has been established. Short nodal segments with axillary buds were isolated from 2-month-old seedlings as culture material. The surface-sterilized explants whose basal ends were soaked in 1, 000ppm of AgNO3 solution for 3 seconds were cultured in (IS IDE and SAITO) medium with different concentrations of BAP (6-benzylaminopurine) or without BAP. BAP concentrations in the media proved to be effective for axillary shoot elongation. Five weeks after the inoculation of the explants, elongated shoots were cut from them. The basal ends of the shoots were soaked in STS (silver tiosulfate) solution for 5 seconds. The shoots were transplanted to the rooting medium which was modified from IS medium. A rooting rate of 44.4% was achieved 7 weeks after the transplanting.
