抄録

The present study was designed to develop a simple and quantitative technique evaluating the extent of freeze damage in a fowl spermatozoon. Acrosin proteolytic activity (APA) by both of non-frozen and frozen-thawed spermatozoa in glucose solution containing various levels of glycerol was measured using a gelatin substrate slide. The mean diameter of 100 halos made by spermatozoal digestion was used as an indication of APA. There was no significant difference in the APA of non-frozen spermatozoa among any groups. After freeze-thawing, the APAs of spermatozoa in all groups decreased remarkably as compared with its of non-frozen spermatozoa. The APA of frozen spermatozoa significantly increased with the increase of glycerol concentration from 0 to 7%, but they decreased significantly according to the increase of glycerol concentration beyond 10%. Therefore, this technique appears to be a more effective and more simplified method for evaluation of freeze-thaw damage of the acrosome in fowl spermatozoa.
