This study was performed to know the fluctuations of luteinizing hormone (LH) and prolactin (PRL) syntheses in the anterior pituitary gland and releases of these hormones from the anterior pituitary during ovulatory and nonovulatory days in hens. The anterior pituitary gland and blood samples were collected at various times during the ovulatory cycle of laying hens or the corresponding period of nonlaying hens. The LH βsubunit and PRL mRNA concentrations in the anterior pituitary gland were measured by solution hybridizationRNase protection assay, which is highly sensitive and quantitative. In addition, LH and PRL concentrations in the anterior pituitary and serum were measured by radioimmunoassay. In laying hens, a preovulatory peak in the serum LH concentration was found 6-7 hours before ovulation. Maximum peaks of the LH-, 6subunit mRNA and LH concentrations in the anterior pituitary gland preceded by three hours before the preovulatory peak of serum LH. In nonlaying hens, no changes in the concentrations of PRL mRNA and PRL in the anterior pituitary were found during nonovulatory days. However, both laying and nonlaying hens showed peaks of serum PRL at 0400-0500hr. Also, the anterior pituitary PRL mRNA concentrations, as well as the pituitary and serum concentrations of PRL, were higher in laying hens than in nonlaying hens in all periods. These results indicate that the synthesis of LH in the anterior pituitary is stimulated before the preovulatory release of LH, they suggest that in laying and non laying hens the release of PRL from the anterior pituitary is controlled by a photoperiod, and that PRL may participate in egg-laying.
