The effects of sperm concentration on the glucose utilization of cock spermatozoa were investigated. The spermatozoa were washed twice with phosphate buffered solution containing 0.02 M glucose. The washed spermatozoa were resuspended at different concentrations in the phosphate buffered solution containing various levels of seminal plasma, blood serum, Lake's B solution and Ringer solution. The sperm suspension was incubated in the water bath with a shaker at 41 °C for 2-4 hours. The glucose concentration of each sample was measured before and after incubation.<br>The results obtained are as follows.<br>1) When the washed spermatozoa were suspended in phosphate buffered solution, an increase in sperm concentration resulted in a comparative increase in glucose utilization per 108 spermatozoa.<br>2) The glucose utilization per 108 cells was closely related to the concentration of spermatozoa in the media containing 15% seminal plasma or blood serum. However, when the media contained 50 and 83% seminal plasma or blood serum, the glucose utilization per 108 cells declined with increasing sperm concentration within the ranges of its concentration studied.<br>3) When Lake's B or Ringer solution was contained in the media, an increase in sperm concentration was accompanied by an increase in the glucose utilization per 108 cells.<br>4) The glucose utilization of spermatozoa was fairly increased by addition of large amounts of seminal plasma or blood serum.<br>5) These results show that seminal plasma and blood serum contain some factor (s) stimulating glucose utilization of spermatozoa and that a decrease in sperm concentration stimulates the glucose utilization of cock spermatozoa in the media containing large amounts of each body fluid. This suggests that transparent fluid and the reduction of sperm concentration by addition of its fluid cause a stimulatory effect on the glucose utilization of cock spermatozoa at natural copulation.
