Abstract

Yolk sphere formation relating to the follicular transformation was compared histochemically among small follicles and yellow growing follicles in hen's ovary. The hens laying an intraclutch egg (Cs) from 09:00 to 10:50 on the experimental day were killed 2, 8 and 14h after the oviposition. Yellow growing follicles and 5 to 6 small follicles were collected from the ovaries. Cryostat sections of these follicles were stained with Baker's acid hematin test to examine the yolk sphere formation.<br>White yolk sphere layers were discontinuously formed in prehierarchical small follicles, while yellow yolk spheres were present in the yolk of yellow growing follicles. It was noted that one small follicle exhibiting the formation of yellow yolk spheres near the perivitellin membrane was found in all the ovaries from hens killed at 14h after the oviposition, corresponding to the time shortly after the occurrence of follicular transformation. The present results show that observation on the formation of yellow yolk spheres is useful as a criterion to judge the follicular transformation.
