Abstract

Negative ions from neutral oligosaccharides were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The deprotonated molecule [M−H]- were decomposed in negative-ion MALDI using nor-harmane as a matrix, indicated that the in-source decay took place at the laser irradiation. The chloride ion adducts [M+Cl]- were stable enough to be detected by reflectron-mode, and postsource decay (PSD) spectra were measured from those ions. The PSD ions were detected as the deprotonated ion form rather than the chlorinated one, indicated that the deprotonated molecules were very labile and a reaction intermediate in the PSD. The in-source decay ions depended on the linkage types of oligosaccharides and their spectra could be useful to identify the linkage types. The PSD spectra were also useful.
