Electron spectroscopy for chemical analysis studies on polypeptide surfaces.

Abstract

The outer-most surface characteristic of poly (γ-benzyl L-glutamate) (PBLG) and poly (ε-N-benzyloxycarbonyl L-lysinc) (PBCL) were investigated by electron spectroscopy for chemical analysis (ESCA). These homopolypeptides exist in the α-helicai conformation in the solid state. The ESCA measurements showed the ratios (O/C) of atomic concentrations of both polypeptides are in accord with the bulk values for grazing angles from 45°C to 60°C. As the result of the evaluation of effective sampling depth estimated from the electron mean free path, we obtained d=16.3Å to 19.9Å for PBLG and d= 16.6Å to 20.4Å for PBCL in the same grazing angle range. The diameters of the cross-section, a, of PBLG and PBCL helical rods have been reported as a=15.0Å and 18.0Å, respectively. Agreement of d- and a-values for PBLG and PBCL seems to be satisfactory. These results suggest that the depth profile of polypcptide surfaces existing in the α-helical conformation is specific and the effective sampling depths of both PBLG and PBCL surfaces are in accord with the diameters of the cross-section of their helical rods over the same range of grazing angles.
