Description

Molecular weights of chitosan samples, which were obtained by alkali treatment of crab chitin, were determined by a GPL/LALLS method. The degree of deacetylation of the chitosan samples used was in the range from 78 to 99%. The weight-average molecular weights (Mw) of the samples ranged from 8.5×105 to 4.9×105 g/mol, the molecular-weight distribution (MWD) being from 1.55 to 2.08. The Mw values decreased with the rise of treatment-temperature and with the increase of deacetylation, giving rise to larger MWD's. Degradation of chitosan in aqueous acetic acid solution was investigated. The decrease of Mw was only 5-20% even after 80 days when it was stored at 4°C, the decrease being smaller for more deacetylated samples. Film preparation from the aqueous acetic acid solution did not cause any decrease of Mw. When chitosan films were stored in water, the molecular weight significantly increased for the sample of 99% deacetylation, but the films became insoluble if the deacetylation was 87%.
