Description

This review contains the historical development on the insulin structure study which was begun in 1935 by D.C. Hodgkin, and the relationship between the structure and the biological activity. As 2 Zn insulin crystals belong to space group R3 and the positions of the main heavy atom in the most derivatives are very close to the three fold axis, many difficalties had to be overcome during the structure determination. We describe the three dimensional arrangement of atoms in the molecule in 2 Zn insulin crystal and particularly its states of aggregation. The binding site of insulin receptor is proposed on the basis of the three dimensional structure, biochemical studies, and the studies using modified insulin, and the structure of proinsulin is also mentioned.
