<p>The bone marrow toxicity of doxorubicin (DXR) encapsulated in polyethylene glycol-liposome (DXR-PEG-Liposome) in rabbits was examined and compared with DXR encapsulated in conventional liposome (DXR-Liposome) and free DXR. The white cell counts and platelet counts were measured for 28 days following intravenous injection at a dose of 1, 3, or 5 mg/kg. At that time, serum lipid peroxides (LPO), creatine phosphokinase (CK-MB), body weight, and temperature were also measured. The myelotoxic level of DXR-PEG-Liposome as judged by white cell counts and platelet counts was less than that of free DXR. However, no significant differences were found between DXR-PEG-Liposome and DXR-Liposome. It was suggested that such low toxicity of DXR-PEG-Liposome is due to a low accumulation of DXR in bone marrow. The changes in LPO and CK-MB were not observed; neither were the losing weight and fever, even at high dose (5 mg/kg). These results suggest that DXR-PEG-Liposome has little toxicity for the bone marrow in clinical application.</p>
