Acetylcholinesterase (AChE) inhibitory activity of acetone extract from house dust was investigated. AChE activity was measured by using acetylthiocholine iodide as a substrate and colorimetry by 5,5'-dithiobis(2-nitrobenzoic acid)(DTNB). House dust was obtained from ordinary houses around Osaka in eight samples in 2013, 8 in 2014, 15 in 2015, and 20 in 2016. A sample for measuring the enzyme inhibitory activity was extracted with acetone from house dust under ultrasonic wave. After acetone was removed from the extract, the residue was dissolved in ethanol. AChE inhibitory activity was measured as follows: mixed AChE and the extract from house dust in a quartz cell, added phosphate buffer and DTNB, allowed to stand for 5 minutes, added a substrate, and then measured the absorbance at 412 nm up to 5 minutes. The enzyme inhibition rate of the sample was showed as the ratio of increasing rate of absorbances between the samples and ethanol as a control. All of the extracts from house dust samples showed AChE inhibitory activity (inhibition ratio 7-79%). When eight kinds of organophosphorus flame retardants (PFRs) were measured for the inhibitory activity by the same method, the inhibitory activity was observed in all of them. Several kinds of PFRs were detected in all of the extract from house dust samples.
