Abstract

<p>We developed a new method of intratympanic (ITy) dosing involving puncture of the eardrum in male guinea pigs and evaluated the effects on the auditory system by auditory brainstem response (ABR), histopathology, and scanning electron microscopy. In the 10-day repeated dose (100 μL/ear), saline and 4% gentamicin showed elevated ABR thresholds (ABRt); furthermore, gentamicin showed a loss of inner and outer hair cells in the ductus cochlearis. In the weekly dose (4 times) of saline, no ABRt changes were noted. From these results, ITy dosing is useful for ototoxicity evaluation in guinea pigs.</p>
