Biodegradable porous microneedles via PLA microspheres for rapid ISF extraction

<p>This study presents a novel fabrication method of biodegradable porous microneedles (MNs) for pain-free interstitial fluid (ISF) extraction beyond the skin. Biodegradable polylactic acid (PLA) microspheres prepared by single emulsion were used for MNs material. Microspheres were filled into a mold in order to form continuous capillaries directly and then heat treatment was implemented to bond microspheres together to produce interconnected micropores and robust porous structures. The results demonstrated that the extraction performance of MNs improved greatly after the heat treatment above the melting point of PLA. In addition, the determination of glucose level was confirmed within 1 minute by using a paper-based colorimetric biosensor using fabricated MN array. We expect the proposed biodegradable porous MNs enable the scale-up and mass production by its simple and low-cost fabrication method. Furthermore, the porous MN array integrated with biosensors showed the potential to realize minimally invasive, early and rapid diagnosis of diseases. </p>
