Abstract
<p>The electrorotation (ROT) rates of K562 cells accompanied by erythroid differentiation were measured to identify the differentiation status induced by three kinds of agents, which were butyric acid (BA), nicotinic acid (NA), and isonicotinic acid (INA). For induction of ROT, a novel electrorotation device with a microwell arranged on a center of polynomial electrode was fabricated. The polynomial electrode consisted of four electrodes arranged symmetrically around a point. The single K562 cell was trapped in the microwell and was subjected to ROT. The ROT rate increased with the differentiation of K562 cells, because the cytoplasm conductivity would increase with an increase of the concentration of iron ions to produce hemoglobin. The ROT rate could be utilized to estimate the stage of cell differentiation without labeling.</p>
