Abstract
<p>In the present work, facultative anaerobic bacteria, Escherichia coli (E. coli), were straightforwardly immobilized into the conducting polypyrrole (PPy) by electrochemical deposition. PPy plays an important role as a substrate for entrapping bacteria because of the resulting density, viability, and distribution. The proliferative process of E. coli cells on the PPy film was monitored by using a dark-field microscope. The cell density of E. coli on the PPy film, which was counted as 5.2 × 105 cells cm−2 increased logarithmically to 1.4 × 107 cells cm−2 after incubation for 18 h under aerobic condition. During, incubation, the formation and growth of biofilm were observated clearly on the PPy film. Electrochemical cultivation promoted bacterial growth under the anaerobic condition, since an applying potential of +1.0 V (vs. Ag|AgCl) makes it possible to supply nitrate continuously by an oxidiation reaction of nitrite to nitrate in a liquid medium. The respiratory activity of bacteria on the PPy film was clearly observed regardless of the anaerobic and aerobic conditions, suggesting that this platform can serve as a simple, but powerful, analytical tool to monitor bacterial activity.</p>
