Abstract
In this study, we investigated the effect of ovarian hormones on the thickness of human endometrial stromal cell sheets in vitro. The endometrium gets thickened due to the influence of ovarian hormones with the menstrual cycle, which allows implantation of a fertilized egg. However, infertility patients sometimes do not experience this thickening, and thus drug has been developed is required. Therefore, the efficiency of drug development could be improved by using cell sheet technology to construct a three-dimensional tissue that exhibits the same hormone response as in vivo. In this study, we examined the harvesting conditions of human endometrial stromal cell sheets and analyzed the change of cell sheets thickness by adding hormone. As a result, the thickness of the cell sheet cultured in the culture medium containing the hormone was about 1.5 times compared with no hormone. The present results show that human endometrial stromal cells change their morphology under the influence of ovarian hormone even in vitro, suggesting that they can be used for the preparation of a model of endometrial mimicry.
