A multi-functional organelle, mitochondrion, participates in a wide variety of cellular processes from energy metabolism to cell death. Current researches on mitochondria in mammals revealed that they involve in the cellular innate immune response against infectious microorganisms or sterile activators. The mitochondrion-related innate immune response is accomplished by the activation of two distinct signaling transduction pathways, that are the retinoic acid-inducible gene I (RIG-I) pathway and the NLR family pyrin domain-containing 3 (NLRP3) inflammasomes. In both signaling pathways, a mitochondrial outer membrane adaptor protein, mitochondrial antiviral signaling (MAVS), acts as a key role in the signal transduction. In this review, we take a view of our current insights regarding the fundamental phenomena of mitochondrion-related innate immunity, and review the specific roles of the mitochondrial zone responsible for the immune responses.
