The effect of external Na+ removal on ACh contracture and catch tension in intact ABRM and on the contraction and relaxation in chemically skinned ABRM, prepared by the treatment with EDTA, of Mytilus was studied, and the following results were obtained. 1) Contracture tension induced by ACh reduced with time when all the external Na+ was removed by the replacement with isosmotic sucrose, and was abolished within about 60 sec. Even in the case in which Na+ removal and the application of ACh was carried out simultaneously, the contracture tension was reduced by about 30%. 2) On the other hand, the catch tension after ACh contracture commenced to be released 15?90 sec after the Na+ removal, and was completely released after 3?4min. 3) Chemically skinned ABRM developed tension when it was immersed in high (10-5M) Ca2+ solution and relaxed in low (10-9M) Ca2+ solution containing 5mM ATP, 5mM MgCl2 and 125 or 250mM KCl. The tension in high Ca2+ solution was reduced when KC1 was increased to 450mM. 4) The rate of relaxation in low Ca2+ solution decreased with the period of the preceding immersion in high Ca2+ solution, and when the immersion period was 12?15 min, the rate decreased to 2/3?1/2 times that in the case where the immersion period was 3?5min. 5) No difference between the effect of 125 or 250mM KCl and of isosmotic NaCl on the contraction and the relaxation in high and low Ca2+ solution was observed. 6) The rate of relaxation under these conditions was not altered in most cases by 10-4 M 5-HT, but increased when 0.5 mM theophylline was added to 5-HT.
