Occupational stress has generally increased in Japanese workers in recent years. To prevent stress-related diseases and to promote individual and occupational health, the assessment of stress levels by means of an objective and quantitative method is desirable. It has been suggested that salivary alpha-amylase (sAMY) reflects sympathetic activity and is a useful stress-related biomarker, because saliva collection is easy, stress-free and noninvasive. A rapid and easy-to-use hand-held monitor (Amylase monitor, Nipro Co., Ltd.) for the measurement of sAMY activity has also been developed. However, more fundamental studies regarding several kinds of variations of sAMY need to be examined, when we discuss the availability of this device in relation to occupational stress. To evaluate these issues, we investigated diurnal, weekly and seasonal variations of sAMY acivity at the workplace in a total of 17 office workers. The levels of sAMY appeared to show slight elevations on Monday in comparison with other weekdays and around 10 AM during daytime on Monday, Wednesday and Friday, however, no significant differences were found. The values of sAMY seemed to indicate slight elevations during winter and spring, but, no significant differences were also observed. The suggestions of diurnal, weekly and seasonal variations of sAMY acivity at the workplace are not confirmed in the present study and remain to be elucidated. Further studies on the basis of larger samples are required to clarify our suggestions.
