雄性両性異株マルバアオダモの花粉の発芽特性を調べるために、ホールスライドHanging Drop培養法、吸水セロハン・ショ糖培地方、ショ糖・寒天培地法での花粉培養液を用いた発芽率を調べた。ホールスライドHanging Drop培養法では、pH、ショ糖濃度を変化させて実験を行ったが実験条件下は発芽がみられなかった。吸水せろはん・ショ糖培地(pH6、ショ糖濃度10%)でも花粉発芽は確認されなかったが、この培地に雌しべの抽出物を転化した場合には発芽が見られた。ショ糖・寒天培地では、10%前後の低い発芽率が観察された。発芽率は雄株花粉が両性花株花粉のものよりも高い値を示したが、統計的には有意ではなかった。一方、蛍光顕微鏡下での受粉花粉の観察では30%以上の高い発芽率が観察され、この植物の発芽観察には直接観察による比較が必要であることが判明した。また、6時間後には、花粉管は花柱中ほどまで伸張していたが、まだ胚珠には到達していなかった。24時間後には胚珠にまで到達していることが観察された。伸張した花粉管は柱頭上では多数みられるにもかかわらず、花中の中部では1～2本しか確認されず、花柱上部で花粉管競争が起こっていることが示された。

The pollen germination of the androdioecious tree, Fraxinus sieboldiana was investigated in three kinds of culture methods; the hanging drop culture, the culture on the cellophane menbrane and the culture on the afarose surface. The effects of pH (6,7 and 8) and sucrose (0%, 10%, 20%) were studied in the hanging drop culture. No people germination were observed in this culture. There was also no pollen germination in the culture on the cellophane membrane, but less than 15% germination were observed in both the culture added with the pistil on the cellophane membrane and the culture on the agarose surface. The pollen germination percintage of male flower was larfer than those of hermaphrodite flowers in these cultural conditions, however, there were not statistically significantly differences on the pollen germinaiton between sexes. Direct observation of the pollen germination on the stigma under the fluorescent microscope, stained with the aniline blue fluorescence method showed more than 30% of the pollen grains germinated in vivo in both sexes. The pollen tubes did not reached the ovule in 6 hours after the hand-pollination, and only one or two pollen tube elongated toward the mid part of style. Only one pollen tube reached the ovule after 24 hourses. This indicates that there are pollen tube competitions between the stigma and the mid part of style.
