Description
The transformation of a recalcitrant cultivar of chrysanthemum, Dendranthema grandiflorum Kitamura cv. Prelude-coral, was conducted using petals as explants infected with Agrobacterium tumefaciens harboring a Ti-plasmid containing neomycin phosphotransferase (NPTII) and β-Glucuronidase (GUS) reporter genes. To reduce the number of false-positive calli, a method that the explants are soaked in a medium containing paromomycin (the selection agent for transformed cells) was developed. Seven putative transgenic plants were regenerated on the selection medium from 875 explants (efficiency 0.8%), and 6 of them were devoted to molecular analyses. PCR-based analyses of genomic DNA indicated that all the plants contained the NPTII gene, but 2 plants were missing the GUS gene. A reverse transcription PCR (RT-PCR) analysis showed that the mRNA of NPTII was expressed in 6 plants. On the other hand, only one plant expressed GUS mRNA, but no GUS enzyme activity.
