Abstract
<p>Vialinin A having the 2’,3’,4,4”-p-terphenyltetraol moiety exhibited potent inhibitory activity against immunologically stimulated histamine and TNF-α release from a human cronic myelogenous basophilic leukemia cell line, KU-812 with IC50 values of 30.0±2.8nM and 0.04±0.01nM respectively. In addition, a vialinin A-binding molecule in KU-812 was isolated by using a biotinylated bioprobe and identified to be Ubiquitin-specific peptidase 5 (USP5)/isopeptidase T (Iso T), binding specifically to 5’,6’-dimethyl-1,1’,4’,1”-terphenyl-2’,3’,4,4”- tetraol (DMT) moiety, by peptide mass fingerprinting method. These results show that the inhibitory mechanism of vialinin A against TNF-α release from KU-812 is similar to that from a rat basophilic cell line, RBL-2H3. And vialinin A would be applied to therapeutic agents, based on a novel model of actin, targeting inflammatory affections induced by TNF-α such as rheumatoid arthritis and Crohn’s disease.</p>
