A new quantitative method of secretory IgA (referred to as S-IgA) and free secretory component (referred to as FSC) was investigated by Mancini's single radial immunocliffusion (SRID) method using anti-secretory component (referred to as anti-SC) serum. This antiserum was obtained by immunizing rabbits with FSC emulsified with complete Freund's adjuvant, which was allowed to react with both FSC and bound SC (referred to as BSC) in the S-IgA mocleule and the results showed an identical precipitin line on Ouchterlony plate and immunoelectrophoresis. However, differential double precipitin rings of FSC and BSC in S-IgA molecule were noted in the reaction by SRID. Linear calibration lines were drawn from the SRID precipitations between anti-SC and FSC and S-IgA of various concentrations, respectively. In practice, differential and simultaneous quantitation of S-IgA and FSC were possible in exocrine secretions such as saliva and gastric juice.
