Onion yellows phytoplasma (Candidatus Phytoplasma asteris) is a phytopathogenic bacterium that causes witches' broom, yellowing and stunting in plants. Ca. P. asteris OY-W strain and OY-M strain induces different symptoms to infected plants from each other. OY-W strain induces severe symptoms such as witches' broom, stunting and yellowing in plants, whereas OY-M induces only mild symptoms of proliferation. It has been suggested that glycolytic genes may be involved in the cause of this difference in virulence because glycolytic genes are single-copy on the genome of OY-M strain, whereas OY-W strain has two copies of those genes. In OY-W strain, there is a possibility that the duplication of genes enhances the function of a glycolysis pathway, thus large amount of glucose is consumed in the host cells, leading to severe symptoms. However, it has not been clarified whether both two copies of glycolytic genes have normal functions in OY-W strain. In this study, I aimed to investigate the enzymatic activity of phosphofructokinase which is the rate-limiting enzyme of glycolytic pathway, by cloning two pfkA genes from the OY-W strain.
