Experiments were carried out to solve the question as to why the ovaries of hypophysectomized female rats united parabiotically with gonadectomized partners seldom develop corpora lutea. Hypophysectomized-gonadectomized female rats were given either intrasplenic or subcutaneous ovarian grafts and united parabiotically with gonadectomized male or female bearing intrasplenic ovarian grafts. At sacrifice 4 weeks after union, intrasplenic grafts in gonadectomized partners had many corpora lutea in addition to follicles. In hypophysectomized-gonadectomized parabionts, subcutaneous grafts exhibited many large and small follicles but no corpora lutea, while intrasplenic grafts showed some corpora lutea as well as many follicles. These results can be explained as follows. Subcutaneous ovarian grafts in hypophysectomized parabionts secrete hormones under the influence of gonadotrophins coming from gonadectomized partners with their hypophysis intact. The ovarian hormones, entering the blood of gonadectomized partners at a concentration lower than that in their co-twins, act on their hypothalamo-hypophyseal system and decrease the secretion of gonadotrophins from the hypophysis. The hormones in turn enter hypophysectomized-gonadectomized parabionts at a more decreased level insufficient to cause luteinization in their subcutaneous ovarian grafts. In contrast with this, if both parabionts have intrasplenic ovarian grafts, the hormones secreted by the grafts exert no feedback action on the hypothalamo-hypophyseal system of the parabionts with their hypophysis. Consequently gonadotrophins are secreted by the hypophysis of these animals without any suppression, and transferred to hypophysectomized-gonadectomized parabionts at a lowered concentration which can still induce both follicular growth and luteinization in their intrasplenic ovarian grafts.
